Subject: frozen WL yeast vial - still ok?
|
Hello All We've had a bit of flooding locally and I was without power for 3+ days. I pulled a vial of WL ale yeast from the (warming) fridge and put it in the freezer hoping to hang on to it. The power kicked in over night, the freezer kicked in, and now I have a frozen WL vial. Is the vial still ok? I always make a starter anyways, but if it's possibly b0rked I can pick up a new vial at the LHBS today. Got within a 1/4 mile of it and it seemed to be up high enough to not be overrun, but I can't be sure, couldn't see it that clearly. Phone is dead, too :-(. Doc's Homebrew in Binghamton, NY. I hope Doc makes it. ( insert moment of silence ) In any event, I'm going to try that WL vial in a starter this afternoon.. ----------------------------------------------- John Bleichert syborg@earthlink.net The heat from below can burn your eyes out!!

Freezing will kill some of the Yeast, but some of them will survive. Make a starter a couple of days earlier that you would normally. If it takes off fine, if not, you will still have time to get another vial. IE, was it just slushy or was it a solid block of ice. When the water that is absorbed inside the yeast cell freezes, it crystalizes and expands, often rupturing the cell. The yeast doesn't tend to like that. > Is the vial still ok? I always make a starter anyways, but if it's > possibly b0rked I can pick up a new vial at the LHBS today. IMO, I'd definitely make a starter first to make sure it's alright, but there's a posibility that it's bOrked. John.

Streak them onto a peatry dish and see if you can get any colonies to grow. If you do, you can make a strter from just a few colonies. If you don't have any petri dishes you can order them from www.morebeer.com. http://morebeer.com/browse.html?category_id=1153&keyword=&x=1&y=1 For the media use gelatin and DME. Once cultured you can store them in your fridge for 4-5 months before they get ugly, then all you have to do is pluck off a colony and re-colonize a new petri dish. I use to have 3-4 yeast strains on hand in petri dishes in the fridge. The wife hated it, but...

Here is a REALLY good article on yeast culturing: http://hbd.org/brewery/library/YeacultBL.html
